First, this is not a ZQuest problem. Moving to the correct forum...
Second, go here. The procedure described here worked for the person asking, so it can work for you, too. :)

Are you trying to play a quest, or load a quest to work on? If you want to play a quest go to Zelda-w(for beta 163) or ZeldaClassic(original) and load a custom quest, if you want to work on a quest, either import graphics by file-import-graphics pack-file name, or file-open-quest name. Make sure your doing this part in Zquest.
