Yes, it's simple. Place a closed treasure chest on the screen. Put flag #10 Armos (->Item) on it. Set the undercombo as the opened treasure chest, then set the room type as Special Item and the special item as whatever you want. You should also go to Data -> Screen Data and check Hold Up Item. That's it!

Just keep in mind that if you use the undercombo for the unlocked chest then all the undercombos on that screen will be the open chest. You may have to play with warps if you plan on doing something more complicated than just having the chest in the room with some enemies.

Multiple undercombos can be used, via Secret Combos 16-31. Just put flag 16 over the chest and then go to secret combos. Have #16 set to the open chest. It should work...

You don't have to use the locked treasure chests.

There are 6 new combo types:

Treasure chest
Treasure chest (Copycat)
Treasure chest (Locked)
Treasure chest (Locked, Copycat)
Treasure chest (Boss)
Treasure chest (Boss, Copycat)
This way, there is no need for Under Combos or Seceret Combos. (the open Treasure chest is always the next one in the list.)
For a small Treasure chest, set the combo type to Treasure chest and the flag to Armos->Item.
For a large Treasure chest, (the same flag on all for Combos) set the bottom two combo types to Treasure chest and the top two to Treasure chest (Copycat).
