Is it possible to make a tile that will damage you when you walk on it, but you can't walk past it? I would like to use such a tile in my quest, but I'm not sure how to get it...

So you mean a solid damage combo? Unfortunately, this cannot be done. Maybe some expert has a crazy way to do it, but it's out of my knowledge. But I'm very fairly certain it isn't possible. :(

Oh well. I guess there's other ways, like putting solid blocks in places so you HAVE to follow the safe path to get out...

Or you can put unwalkables after said tiles.

An idea was sparked by rocksfan13... You could even make the innermost part of the damage tiles not solid, and the outermost part solid so you can only walk on a little of the tile to get hurt.

Say this is the room you want to make, where each character represents a combo, and each different kind of character is a different combo.

9 7 7 7 7 7 8
5 E E E E E 6
5 E E E E E 6
5 E E E E E 6
4 3 E E E 2 4
4 4 3 E 2 4 4

Let's say you want to walk on "E" without getting hurt. Any other character (2, 3, 4, 5, 6, 7, 8, 9) is a damage combo. (You should know that the NW, NE, SW, or SE quarter of the combo can be solid or not.)

E - entire combo is not solid
2 - SE of combo is solid
3 - SW of combo is solid
4 - entire thing is solid
5 - NW, SW of combo is solid
6 - NE, SE of combo is solid
7 - NW, NE of combo is solid
8 - NW, NE, SE of combo is solid
9 - SW, NW, NE of combo is solid

Therefore, only any quarter of a combo touching "E" is walkable and is damaging. Anything that is NOT touching "E" is solid.

Confusing, yes. Works? Yes.
